先天免疫是宿主抵御病毒感染的第一道防线。 病毒成分(尤其是其基因组或复制中间体)会被不同的模式识别受体(PRRs)感知,包括Toll样受体(TLRs)、RIG-I样受体(RLRs)/或细胞内DNA传感器。PRRs对病毒核酸的感知会触发信号级联,从而诱导I型干扰素(IFN)、促炎细胞因子和其他下游抗病毒效应基因的转录。下游的细胞因子和效应物抑制病毒复制或诱导受感染细胞的凋亡,导致先天性抗病毒反应。但是体液代谢是否以及如何调节先天抗病毒反应仍然是个谜。最近,武汉大学中南医院医学研究所免疫与代谢前沿科学中心在期刊《Signal Transduction and Targeted Therapy》(IF=18.187)上发表题为“Modulation of innate immune response to viruses including SARS-CoV-2 by progesterone.”的文章,发现病毒感染通过小鼠下丘脑-垂体-肾上腺轴诱导孕酮。孕酮诱导下游抗病毒基因并促进细胞和小鼠的先天抗病毒反应,而敲除孕酮受体PGR具有相反的效果。孕酮刺激PGR激活酪氨酸激酶SRC,使转录因子IRF3Y107磷酸化,导致其激活并诱导抗病毒基因。感染新型冠状病毒病毒的患者孕酮水平升高,这与新冠肺炎的严重程度降低相关。该发现揭示了黄体酮如何调节宿主先天抗病毒反应,并指出黄体酮是一种潜在的感染性和炎性疾病的免疫调节剂。
