肺转移是癌细胞从我们身体其他地方发展的原发性肿瘤扩散到肺,这被认为是癌症死亡的关键原因。其治疗受到转移性肿瘤中有限的药物递送效率和免疫抑制的限制。最近,中国科学院上海药物研究所的研究团队在期刊《ACS nano》(IF=18.027)上发表题为 “Spatial-Drug-Laden Protease-Activatable M1 Macrophage System Targets Lung Metastasis and Potentiates Antitumor Immunity.” 的文章,该研究设计了一个空间药物加载的M1巨噬细胞系统,其内部是脂质体R848,在M1巨噬细胞(RDM)的膜上是成纤维细胞活化蛋白蛋白酶(FAP)敏感的磷脂-DM4缀合物。RDM可优先积聚在肺转移病灶处,并作为游离药物分子或载药纳米囊泡响应性地释放治疗剂。与阴性对照组相比,RDM治疗显著增强了CD3+ CD8+ T细胞对肺转移的浸润,并分别导致颗粒酶-B-、干扰素-γ和Ki67-阳性亚型增加8.54倍、12.87倍和2.85倍。此外,RDM治疗在4T1模型中产生了90.99%的肺转移抑制,并在三个鼠肺转移模型中显著延长了生存期。该载药体系为靶向肺转移和增强抗肿瘤免疫用于抗转移治疗提供了可行策略。
本文中使用了欣博盛Mouse TNF-α, IL-1β 和IL-6 ELISA kits检测了细胞培养上清中这些细胞因子的含量。
